Design Notes

With 10 AGTC repeats, the GFP sequence is in the +1 frame with respect to the start codon, so GFP should not be transcribed. The number of AGTC repeats was limited by maximum length of synthesized primers.



Source

The tetranucleotide repeat was placed into synthesized primers that bound to the existing part I13401, and then a PCR reaction was run.
